Unbiased genetic screening is a foundational technique that has been greatly advanced by CRISPR technology. While current CRISPR screens utilize pooled guide RNA libraries, which are limited to selectable phenotypes, arrayed CRISPR libraries significantly expand their scope. However, arrayed CRISPR libraries remain scarce and are challenging to construct.

Utilizing a newly developed massively parallel plasmid cloning methodology,

𝐃𝐫. π‰π’πšπ§π -𝐀𝐧 𝐘𝐒𝐧 has constructed two genome-wide arrayed libraries for human gene ablation, activation, and epigenetic silencing. Each plasmid encodes an array of four non-overlapping sgRNAs designed to accommodate most human DNA polymorphisms. These new libraries surpass existing resources in both efficiency and versatility across various systems, including induced pluripotent stem cells and organoids. Thus, they unlock vast biological frontiers previously inaccessible and serve as a powerful tool for biomedical research.
